Abstract

Lactic acid bacteria were known has a potential for biopreservative and maintaining the safety of food products. The purpose of this research was to obtain lactic acid bacteria that capable to inhibit harmful bacteria in fisheries products. The stages of the methods were isolation of lactic acid bacteria from the digestive tract tilapia and frigate tuna with MRS agar + CaCO3, antibacterial test against Bacillus subtilis, Morganella morganii, and Escherichia coli using agar well diffusion method and test the alleged strengthening  lactic acid bacteria. The results showed that there were two isolates of lactic acid bacteria that have the greatest inhibitory for harmful bacteria tested.Key words: antibacterial, biopreservative, inhibitor harmful bacteria, lactic acid  bacteria

Abstract

The study about chemical and physical characteristics, bioactive compounds and  antioxidant activity of mangrove api-api (Avicennia marina) leaf have been done.  The structure of leaf tissues was analyzed with microscopic method, chemical compositions were determined by proximate analysis, antioxidant activity was determined by DPPH method, while bioactive compounds were determined by phytochemical assay. The microscopic analysis shown that api-api’s leaf have common feature dicotyl leaves structure, but it has a unique additional structure, that was salt extruding gland.  The result of proximate analysis showed that api-api’s leaf was high in crude fiber (4.12%) and carbohydrate (23.00%).  Ethyl acetate extract of api-api’s leaf showed higher antioxidant activity than hexane and methanol extracts, with IC value is 182.33 ppm.  The ethyl acetate extract in concentration 300 ppm had higher ability to inhibite autooxidation that occurred in oil/water emulsion.  Flavonoid, steroid dan reduction sugar were detected in api-api leaf extract.Keywords: antioxidant activity, Avicennia marina, DPPH, flavonoid, salt extruding gland.

Abstract

Proteases constitute one of the most important groups of industrial enzymes, therefore screening of these proteases from bacteria is important. The aim of this study was to characterize proteases from plant swamp bacteria. The result of  proteolytic assays revealed three isolates possesed proteolytic index >1.00 (T1P4, H1P4 and K1P4). The optimum fermentation time of each isolate was 48 hours. The optimum pH of proteases from T1P4, H1P4 and K1P4 was 8.0; 7.5; and 8.0,  respectively. The optimum temperature of T1P4, H1P4 and K1P4 protease was 50 °C; 40 °C;  and 50 °C, respectively. Metal ions (Fe, K, Mn, and Zn) inhibited T1P4 protease except Fe2+ (5 mM), H1P4 protease except  Zn2+ (5 mM),  K1P4 protease except Fe (1 and 5 mM). Effect of inhibtor specific (EDTA) shown EDTA inhibit all  proteases. Study on the effect of metals ion and spesific inhibitors indicated that all protease were  metaloprotease. The moleculer weights of T1P4, H1P4 and K1P4 proteases were determinited by using SDS-PAGE and zymography technique were 151; 138; and 127 kD, respectively.Key words:  bacteria, characterization, protease, swamp

Abstract

Tujuan dari penelitian ini adalah untuk mengetahui peningkatan prestasi Lari 50 Meter melalui Metode Progresif pada siswa Kelas V SDN Arjowinangun Kecamatan Pacitan Kabupaten Pacitan Semester I Tahun Pelajaran 2017/2018. Pengajaran Pendidikan Jasmani Olahraga dan Kesehatan yang diharapkan adalah pengajaran yang dapat membuat siswa benar-benar mampu menerapkan, bukan hanya menguasai teori saja. Pada kenyataannya pada mata pelajaran Pendidikan Jasmani Olahraga dan Kesehatan, siswa belum dapat menerapkan secara maksimal. Berdasarkan data yang diperoleh peneliti dari daftar nilai diketahui bahwa keterampilan siswa untuk dalam bidang Pendidikan Jasmani Olahraga dan Kesehatan khususnya pada materi Lari 50 Meter sangat rendah, yakni hanya 54,05% dari jumlah siswa dinyatakan tuntas belajar dan nilai rerata yang dicapai hanya 60,54.  Untuk itu, peneliti menawarkan model pembelajaran Metode Progresif dengan harapan minimal 75% dari jumlah siswa tuntas dalam praktik materi Lari 50 Meter. Penelitian ini merupakan penelitian tindakan kelas yang dilaksanakan dalam 3 siklus yang meliputi perencanaan, pelaksanaan, observasi dan refleksi. Data diambil dengan menggunakan instrumen tes, wawancara, angket dan jurnal. Peranan Metode Progresif dalam meningkatkan prestasi Pendidikan Jasmani Olahraga dan Kesehatan materi ajar Lari 50 Meter ini ditandai adanya peningkatan nilai rerata (Mean Score) mulai dari siklus pertama sampai siklus terakhir, yakni : pada siklus I 71,22; siklus  II 74,59,  dan siklus III 81,62. Selain itu juga ditandai adanya peningkatan prosentasi ketuntasan belajar yaitu pada siklus I hanya 64,86%, siklus II meningkat menjadi 83,78%, pada siklus III terjadi peningkatan mencapai 100,00%. Kenyataan membuktikan bahwa penggunaan Model Pembelajaran Metode Progresif dalam proses pembelajaran dapat meningkatan prestasi Lari 50 Meter pada mata pelajaran Pendidikan Jasmani Olahraga dan Kesehatan.
